Are the machines you're trying to send mail from in 192.168.100.0/24?

Have you tried telnetting to the SMTP port from one of the machines
you're trying to use it from?  If you can't connect, is it a firewall
issue?  if you can connect, try putting through an SMTP session by
hand, see if it gives any error messages.
